Hepatitis C is a more widely popular viral disease and an important impacted global issue of social and economic. About 41 million HCV infection in China, prevalence rate has reached 3.2% of the total population. Moreover, 300 to 400 million new cases happened each year [5]. Hepatitis C chronic pathogenesis is uncertain but there is evidence indicate that immune tolerance is the key factor.PD-1 which was expressed higher in patients with HCV chronic infection plays a very important role in the chronicity of HCV, but its mechanism is not still clear. The transcriptional origin, the first step and the tightly controlled one in gene expression, is essential to learn the expression of a gene. The interaction between a promoter and its transcription factors control is the key point in transcriptional origin, so we decided to study PD-1 promoter to explore the reason that PD-1 is higher expressed in chronic infectious diseases.Finally we got the eight recombinant plasmids containing the full length and the seven truncated bodies of PD-1 promoter respectively. The Luciferease assay showed all of them had the higher transcriptional activity. Among them, one of the truncated bodies (1128bp) is the strongest one, so the core promoter of PD-1 is located in -978bp~+150bp of the gene sequence.According to these results, we intend to further definite the transcription factors binding with PD-1 core promoter to explore their functions in chronic HCV infection, thus to provide a new strategy of the clinical treatment of chronic hepatitis C.
