X-ray energy dispersive microanalysis is a technology of speed development for the past 30 years which can analyze sample scathelessly and with a series of advantages such as simple and speedy to handle,the showing is distinct, the analysis area is small, to achieve location analysis, qualitative analysis and quantitative analysis at once. Especially the energy spectrum analysis combine with Electro Microscopy is the only instrument which can accomplish location analysis, qualitative analysis, quantitative analysis and morphologic observation together.Currently, foreign EDX analysis technology of samples have accomplished the quantitative analysis, while our analysis in biological especially in plants remains in qualitative and semi-quantitative level. Therefore, in the field of plant science quantitative analysis is an important aspect of the future research. Based on the Agarose for matrix to establish block and semi-thin biological sample of Pb, Zn, Cd, Na, K. Get linear regression curve with Hitachi S-3400Nâ…¡scanning electron microscope and EMAX(Horiba). And this method is used to be the Populus nigra L. metal stressed and two habitat salt alkaline root element concentration measurement, results show,1. Each standard sample of good uniformity, it's feasible and credibility to have many scanning spots.2. Half quantitative results are far away from real concentrations, only can be used to compare, the relative concentration datas are not believable.3. Have good linear relationship, R2 themselves are larger than 0.9,4. Compared with block standard sample,semi-thin standard sample's linear relationship is better and accuracy is higher.5. The method of quantitative analysis is applied preliminarily in the determination of relevant elements of Populus nigra L. metal stressed and two habitat salt alkaline root, the conclusion is dependable.In this paper, the quantitative analysis of method in biological samples was investigated, CPS-concentration method of quantitative analysis of EMAX was present first, and in two biological samples were validated, the method in biological research has important value and prospects.
