The pregnancy of mammalian is a complicated process, which involved with embryo implantation, the growth and development of fetal, and the corresponding in physiological and tissue changes of uterus,pregnancy changes with neuro-immuno-endocrine regulation which can be enhanced or inhibited by each other. Our research utilize immunohistochemical SP method and the image analysis to study the distribution, expression variation and correlation of 5-HT, 5-HT1A R, VIP and VIP2 R in different stages of pregnancy. To further studying their physiological functions in uterus provide a useful morphological data.The results are as follows:1. The immunoactive products of 5-HT and 5-HT1A R mainly located in endometrial epithelial cells, the glandular epithelium, vascular endothelial cells, vascular smooth muscle cells, uterine spiral arteries, stromal cells, decidual cells, immune cells and muscle fiber. The expression and variation of 5-HT in endometrium, the density of coloration is strongest in the post-implantation and trimester, and then becomes weaker gradually in the late pregnancy, implantation, preimplantation and post partum. In muscular layer, the trend of 5-HT in pregnancy is the same as endometrium. The expression and variation of 5-HT1A R in rat's uterus during pregnancy . The coloration of preimplantation, implantation, post-implantation gradually become stronger, the strongest coloration was in trimester, and then becomes weaker gradually. The expression of 5-HT and 5-HT1A R in the uterus in different pregnancy periods was found to adapt to some certain rules, 5-HT and 5-HT1AR was involved in the regulation of pregnancy.2. The expression and variation of VIP in endometrium, the density of coloration is strongest in the trimester and late pregnancy, VIP is moderate intensity in the post-implantation, the other periods are weaker. In muscular layer, the strongest coloration of VIP was in Trimester, then it is weaker in late pregnancy, and other periods are is lightest staining. The expression and variation of VIP2R in rat's uterus during pregnancy. In endometrium, the density of coloration is strongest in the post-implantation and trimester, and then becomes weaker gradually in the preimplantation, late pregnancy, post partum and Implantation. In muscular layer, the density of coloration is strongest in the trimester, followed with post-implantation and late pregnancy, then it is weaker in preimplantation and post partum, implantation.is lightest staining.The expression of VIP and VIP2R in the uterus in different pregnancy periods was found to adapt to some certain rules, VIP through VIP2R was involved in the regulation of pregnancy.3. The expression of 5-HT and 5-HT1AR in pregnancy had extremely significant positive correlation. The expression of VIP and VIP2R in pregnancy had positive correlation. The expression of VIP and 5-HT in pregnancy had positive correlation.This explain that VIP and 5-HT had coordination in regulation of the uterus.
