We designed and synthesized three kinds of polymers as gene carriers:PEI derivaties which was modified with hydrophilic groups and hydrophobic groups as lipid-like cationic polymes; cationic polymers contained disulfide bond and novel thiophene derivaties.72 polymers with biodegradable, water-solubility improved, low toxicity were prepared. These polymers which were soluble in water were assessed when B16F10 cells were used as receptor cell and enhanced green fluorescent protein (EGFP) was used as gene reporter. Then we chose four new polymers with high transfection efficiency in these products. These four polymers'transfection efficiency was higher than that of bPEI25kDa and exhibited much lower cytotoxicity. These four materials could condence pDNA into nanoparticles with the diameter 150nm-250nm.Compared with lipid-like polymers, the solution of PEI derivaties modified with hydrophilic groups and hydrophobic groups was improved, and the toxicity was reduced. But thiophene derivaties did not indicate well transfection efficiency. The results showed that these polymers with further modified could be novel low cytotoxicity gene therapeutic agents.
