In the early 20th century the people began to devote using the laser to achieve optical rotation method. In 1986 the emergence of optical tweezers, it sent the optical rotation technology to a new research stage. The advantage of using optical tweezers to study optical rotation lies in its ability to capture and suspend particle no mechanical contacts, avoiding the contact of particle and sample pool underside or other particles, so turn away the resistance and disturb. optical rotation adds the one-dimensional angle operation on the basis of three-dimensional operation, realized the angle operation of particle. Optical rotation technology has been used in many fields as manipulating a single micron, submicron particles, and has broad prospects for development. This thesis include the theory :First of all, Clarify the issue of research background and significance of thesis research, described the characteristics of optical rotation of technologies and application prospects. From the theoretical analysis of the optical rotation needed to optical trap and the nature of crystal particles. It also analyzes several methods to realization of optical rotation, and to achieve optical rotation of particles in the usefulness of several methods were compared. Also described the utilize of optical rotation technique in biology, medicine and other fields.Secondly, from the theoretically analyze the interaction mechanism between polarized light and birefringent crystal particles, in the initial rotate conditions of birefringent crystal particles, increased rotation speed method of particles, the optical rotation theoretical calculation and simulationat of birefringent crystal particles under different optical power level. According to the simulation results, discussed of the impact with the incident light ellipticity, crystal particle radius size, thickness and optical axis orientation etc. parameters of realization optical rotation. Finally, according to theoretical research results, using optical tweezers instrument, choose appropriate experiment parameters on the yeast to optical rotation. also the birefringent crystal particle's optical rotation is studied, the particle rotation frequency in the role of polarized light is measured, and the relation curve between rotation frequency and laser power is drawed. The results of experiment are basically consistent with the theory.
