Objective:Analyzing the expression of nuclear transcription factor KappaB and matrix metalloproteinases-9(MMP-9) in the degenerated intervertebral disc through the disc degenerated model of rabbit.Method:1,Japanese white rabbit(N=15) was used in this study.Anterior annulus fibrosus stabing approach was used in creating the intervertebral disc degeneration(IDD) model.Non-affected lumbar vertebraes were used as the own control.2,Relaxation time,disc height and water-containing of disc were measured through X-ray and magnetic resonance(MRI) at different time point;killed rabbits at different time point,harvested disc samples,observed the general and micro-structure changes through visual study and tissue slice;clarified whether or not the disc degeneration happened and how much it was.3,Detected the expression situation of nuclear transcription factorκB and MMP-9 in degenerated and controlled discs through immunohistochemisty at different point.4,Analysed the immunohistochemisty tissue slice with Image pro plus6.0 soft ware;statistical analysed the acquired data with SPSS 13 system.Result:1,Obvious height loses and osteophyte could be found in X-ray view;T2W view of MRI showed the signal intensity of nucleus pulposus reduced;tissue slice observation showed the annulus fibrosus became chaos and abruption,the cell of nucleus decreased and the matrix of nucleus changed as clew over time. 2,Immunohistochemisty analyzing showed that expression of NF-κB was different at different stage of IDD.At the early stage,it was a strong positive expression.And the positive expression reduced over time and with degeneration aggravating;No MMP-9 expressed in nucleus pulposus.But in end-plate,its expression changed with the degeneration degree as positive correlation.Conclusion:1,Stabing the disc annulus fibrosus is a reliable method to create the IDD model in rabbit.2,NF-κB may have double function in IDD:Firstly,mediate the inflammatory and immune reaction,and induce the IDD;secondly,up-regulate the expression of inhibitors of apoptosis,and inhibit the IDD.3,It's not likely that NF-κB directly up-regulate the expresiion of MMP-9 in IDD.
