As an effective and direct approach of research in all proteins, proteomics represents a novel methodological way to investigate the expression of all proteins of a cell, body fluid or organism, as well as to analyze their structure, function, interaction and modification. After the full genomic sequence of over a hundred kind of model biology had been determined, proteomics has been widely applied in studying regulation of metabolism, genetics and development, physiology and biochemistry, pharmacology and toxicology. At present, proteomics has acquired great progress in the investigation of pathogenesis, the searching for tumor biomarkers and the mechanism of how drug works.Conus snails can excrete conotoxins of more officinal and economic value from marine biology .Up to now, conotoxins are the most complicated peptides famous for their post translational processing. It is very hard to synthesize active conotoxin peptide by chemical synthesis method. Many proteins from Conus snails, such as Protein Disulfide Isomerase, modificatory enzyme ,exert regulative and modificatory functions, which affect synthesis and activity of conotoxin.With the increasingly intense destroyed environment , the resource of Conus was short of supplies. So it was urgent task for protecting the precious protein of conus. Technologies of proteomics were available to research Conus snails venom duct protein in integer protein level, then research the protein related to conotoxin synthesis, furtherly , construct protein library of conus, preserve and develop the resource of Conus .The report was to analyze the total proteins by the widely use technologies including 2-Delectrophoresis,Mass Spectrum , chromatogram analysis and biology informations, so as to lay a good foundation for further research on Proteomics of Conus venom duct.The content of research is below:SDS-PAGE and Tricine-SDS-PAGE experiment were compared in separating low molecular weight peptides to find which way was better. The molecular weights of the total proteins of Conus betulinus Linnaeus venom duct indicated most proteins located in three areas:45-66.2kD,18.4-25 kD and under 14.4kD.The experimental conditions of 2-DE in this study was optimized to achieve better 2-DE figure of total proteins of venom duct of Conus betulinus Linnaeus from sample extraction,hydration way,IEF program setup,IPG strip equilibrium time,gel concentration and so on. Under optimized condition, the better repeatability and resolution of 2-DE figure can be obtained.16 protein spots were analyzed with Peptide finger printing by MALDI-TOF- MS.9 spots were matched and identified according to Mr,pI. and PMF The experiment set up a platform using 2-DE and Mass analysis technologies, which was very available for further proteomics research in the future.2-DE map from three kinds of different size Conus betulinus Linnaeus were compared.The results showed the effective protein spots number by PDQuest, then analysize the their difference. By MALDI-TOF- MS, 5 protein spot was identified.On the basis of MS identification result, the experiments were arranged to separating and purifying PDI enzyme by ROTOFOR,SEC,and so on .Purity of PDI researched to 72%. Then the activity of PDI was evaluated in oxidative folding synthesized peptide.All the research work has laid a solid foundation for the study on Proteomics of Conus venom duct.
