ObjectiveTo explore the development and degeneration of pronephrons, the patterns and characteristics about mesonephrons development and differentiation of amphibian, pronephrons and mesonephrons in Bufo raddei at 11 different incubation stages were observed. Through measuring the expression of TGF-β(TGF-β2,TGF-β3), AQP-2, Bax during development of renal tubules in Bufo raddei explore the role and regulating significance of them.MethodsHistological methods were applied. The expression of TGF-β(TGF-β2,TGF-β3), AQP-2, Bax were measured by immunohistochemistry combined with stereological methods in Bufo raddei kidney at different incubation stages. The expression intensity of them was analyzed by IPP.Results(1) The nephrotome cells in the front pairs of somite of Bufo raddei have formed into sub-sections arrangements at the 18th incubation stage. The pronephrons have been developmenting until 30th incubation stage, then degradating. Some mesonephric vesicles appeared in the vicinity of the anterior of the mesonephric ducts at the 26th stage. At the 28th stage mesonephric tubules took shape. At the 30th stage mesonephric tubules'blind ends invaginated, resulting in primitive renal sacs and glomerulus, mesonephric tubules elongated distinctly and curved back and forth. All mesonephric tissue had produced integrated nephric units during the 32th stages. During the 30th-34th stages pronephric tissue from anterior to posterior of the embryos degenerated in turn. At the 34 stage renal tubules lengthened apparently and folded regularly, which presented morpholotical differentiation of collecting convoluted tubules, proximal convoluted tubules and distal convoluted tubules. The differentiation of collecting convoluted tubules is earlier, which induces the differentiation of proximal and distal convoluted tubules. (2) At 26th-32th stages the epithelial cells of proximal tubules and distal tubules of pronephron appeared TGF-β2 immunoreactivity, at 34th-38th stages the epithelial cells of proximal tubules of mesonephron appeared TGF-β2 immunoreactivity and there is the same thing in a young and an adult Bufo raddei; At 26th-30th stages the epithelial cells of proximal tubules and distal tubules of pronephron appeared TGF-β3 immunoreactivity, at 32th-38th stages the epithelial cells of proximal tubules of mesonephron appeared TGF-β3 immunoreactivity and there is the same thing in a young and an adult Bufo raddei; The epithelial cells of collecting tubules always appeared AQP-2 immunoreactivity at 26th-38th stages and there is the same thing in a young and an adult Bufo raddei; at 26th-34th stages the epithelial cells of proximal tubules and distal tubules appeared Bax immunoreactivity, at 38th stages the epithelial cells of proximal tubules of mesonephron appeared Bax immunoreactivity and there is the same thing in a young and an adult Bufo raddei. Images showed the expression of TGF-β(β2,β3) and Bax first displayed up then down, but expression of AQP-2 always displayed up during kidney development of Bufo raddei.
