CoQ10 is the essential substance to our bodies with non-toxic side effects.For its special physical function,CoQ10 can cure various diseases even cancer.In recent years,CoQ10has been widely used in production of food additive and Health care.But the synthesis efficiency of CoQ10 is still very low and the cost of CoQ10 production is very high.So,there need great effort on the research of CoQ10production to meet the growing demand of CoQ10.In this research,in order to improve photosynthetic bacteria(psb)cell production of CoQ10,we optimized the fermentation medium and fermentation conditions based on the fast determination of CoQ10.At the mean time,we also studied the effects of different additives on the production of CoQ10 by Rhodopseudomonas palustris,and the results of this paper are as following:1.After purification and enrichment experiments,we selected 4 productive strains of CoQ10 named LB01,LB02,RB01 and RB02.The strain LB01 has the highest production of CoQ10 among these 4 strains.After a series of experiments,the strain LB01 has been identified to Rhodopseudomonas palustris.2.In this paper,various preservation methods of Rhodopseudomonas palustris were studied.Comprehensive comparison of these methods,the preservation of solid aerobic slope is a more appropriate preservation method to Rhodopseudomonas palustris after we studied factors of breed time,the cost of preservation,dynamic strain,and laboratory equipment requirement.3.The fermentation medium and fermentation conditions of Rhodopseudomonas palustris were studied in this paper.The optimum medium consisted of Urea0.036%,MgSO4·7H2O0.04%,NaHCO30.3%,NaAc0.21%,KH2PO40.05%,NaCl 0.2%,yeast extract 0.5%,peptone 0.82%and trace element solution 10ml/L.The suitable fermentation conditions of Rhodopseudomonas palustris were:temperature 30℃,initial pH6.5,inoculum size8%,the volume of medium was 150ml in 300ml flask volume,the best fermentation time was 64h,lighting condition is 10cm from distance,and the fermentation methods is oxygen-limited fermentation for 32h after aerobic fermentation for 32h..4.In this paper we also studied the effects of different additives on the production of CoQ10 by Rhodopseudomonas palustris,and the results are as following: add 0.073%para hydroxybenzoic acid(phb) and 0.16%tobacco powder after fermented for 48 hours,add 0.042%isoprene after fermented for 62 hours,add 12% carrot juice and 8%tomato juice in the fermentation medium from the first.In this condition,the production of CoQ10 can be improved greatly to 26.57mg/L,which is 65.03%higher than the original yielding.
