In this paper, the suppression subtractive libraries of Candida shehatae which cultivated under different carbon sources (Xylose & glucose) were constructed and analyzed by using suppression subtractive technology. The main results were as follows:Xylose and glucose were used respectively as different carbon sources for C.shehatae cultivation. The TRIZOL kit method was used to extract total RNA. MRNA Purification Kit was used to purify mRNA. Purified mRNA was used as material to construct the suppression subtractive library by PCR-SelectTM cDNA Subtraction Kit. The suppression subtractive libraries include positive library and reverse library. After identifying the quality of the libraries, the differential fragments of the two libraries were respectively selected for sequencing, among which 1000 EST were from the positive library, and 500 EST were from reverse library.Phred / Phrap / Consed software was used to assemble the measured differential sequence separately. The total Unigenes were blasted against the Genebank Nt Database, Genebank Nr Database and SWlSSPROT Database. According to the obtained genetic information, use Gene Ontology to classify existing genes. Genes were selected for Real-time PCR detection. The results of the Real-time PCR detection were further showed that the quality of the two libraries were good.In this study, a preliminary study of the functional genes was conducted at the transcriptional level. The results of this study can provide a good technical platform for the molecular mechanism of metabolism of xylose-fermentation microorganisms, transcriptional level regulation, recombinant strains and other related researches, and its further research on bioinformatics can provide theoretical guidance on system studying of xylose-fermentation to ethanol, and lay a theoretical foundation for the development of cellulosic ethanol.
