In this study,firstly,the responses of AtBN12(Brassinosteroid-Nitric oxide-regulated-12),AtBN15(Brassinosteroid-Nitric oxide-regulated-15) and AtBN16(Brassinosteroid-Nitric oxide-regulated-16) in Arabidopsis thaliana Col-0 to envirnmental stresses are investigated via Real-Time PCR;sencondly,with application of det2 and bril-5 mutants and the basement of their responses to NO and BR,we analyzed the relationship between BR and NO signal transduction pathway and the fuction of NO in BR signal transduction pathway;finally,we have cloned these genes and constructed GFP fusion subcelluar localization vectors(pMDC43) and overexpressed vectors(pMDC32) in order to invistigate the fuctions of them.The results are as follows:1.AtBN12,AtBN15 and AtBN16 are related to stress resistance。To a certain extent, cold and osmotic stresses can significantly enhance AtBN12 expression compared to other stresses.The expression of AtBN15 are suppressed by cold,hot and salt stresses but that of AtBN16 are upregulated by various strees conditions such as cold,hot, osmotic and salt stress.2.AtBN12 may be regulated by NO and BR or by BR singly.The regulation of AtBN15 by NO is influenced by BR.NO and BR not directly regulate AtBN16 expression,furthermore,the effect of NO on AtBN16 expression depends on BR.3.We have successfully constructed three GFP fusion subcelluar localization vectors (AtBN12-pMDC43,AtBN15-pMDC43,AtBN16-pMDC43) and two overexpressed vectors(AtBN15-pMDC32,AtBN16-pMDC32),and transformed them to Col-0 and det2 respectively.By screening the seeds of transgenic plants,we obtained four transgenic plants with one fused GFP about AtBN12-pMDC43 and three that about AtBN15-pMDC43.
