The research based on the physiological nature of the Bifidobacterium bifidum,Bacillus licheniformis and Clostridiun butyricum.Industrial fermentation medium and cultivante condition which were suitable for the industrialized production of three kinds of bacteria were developed by utilizing cheaper material such as waste molasses,corn steep liquor and so on for developing microbial ecological agentsliving which has higher bacteria number.The growth factor of tomato juice and prebiotics of fructo oligosaccharides were added to shorten the length of time from probiotics to logarithmic growth phase in the process of proliferation.The chief indexes were tested by pilot scale test and feeding experiment.The study also explained the technology and dependablity of three kinds of bacteria microencapsulation.The results indicated the best compounding of fermentation medium was golden apple juice 100 mL/L,waste molasses 17 g/L and corn steep liquor 115 mL/L.The best fermn condition was pH 7.8,cultivante temperature 37℃,inoculation amount 3%and cultivante time 36h.The results of pilot scale test which 6 ton broth was above 108cfu·g-1 and the number of bacterial sludge was above 1012cfu·g-1 were consistent with laboratory data.Daily gain,daily feed intake and feed conversion ratio increased by 14.1%,9.1%and 3.5%respectively in the feeding experiment which three kinds of bacteria were added into pig feed.Three microcapsules were manufactured through three emulsification methods.The most of living bacteria number was No3 microcapsule which was treated with the third emulsification method including bacterial suspension,sodium alginate,calcium chloride and chitosan.The particle size of No3 microcapsule is 10~20μm.The detection results indicated microcapsule viable counts were more than 6.50×109 cfu·g-1,the rate of thalline retention was higher than 87.45%,the viable counts which were treated with gastric acid and bile acid were also more than 106 cfu·g-1 indicating high acid resistance.All microcapsule almost disintegrated after 35 min being treated with artificial intestinal juice.The release rate of enteric-coated was 86.67%.Other characters of No 3 microcapsule were stable relatively. Classic accelerating test verificated three kinds of bacteria's viable counts which were stored 1year under room temperature could keep more than 6.61×108 cfu·g-1.
