Horsetail(Equisetum arvense)is a typical example in Sphenophytina and has special evolutionary position in Pteridophyta.The investigation to the biological characteristic of Horsetail spore germination has significant theoretical and practical value.In this study,we deployed 2D-electrophoresis to analyze the proteome of Horsetail spores during the five stages of germination(mature spores,rehydration spores,double cell spores,germinating spores and spores with prothallus cell).By using the pH4-7 IPG gel strips and 12.5%SDS-PAGE gel,the proteins from these distinct stages were seprated,respectively.After staining by coomassie brilliant blue,1243±47,1247±34,1254±31,1229±57 and 1234±40 protein spots were detected on the gels from each stage.The differentially-expressed proteins during the spore germination were analyzed,respectively 146 protein spots were obviously differentially-expressed. By using MALDI-TOF-MS/MS,114 protein spots(represent 95 proteins)were identified.These proteins involved in 16 function categories such as photosynthesis,energy-respiration, lipid metabolism,amino acid metabolism,secondary metabolism,other metabolism,transcriptional regulation-related,protein synthesis,protein fold,protein turnover, membrane and transport,signal transduction,stress and defence,cell structure,unknown proteins.Representativlly,These proteins involved in photosynthesis(17%),protein metabolism(32%)and energy-respiration(14%)were over-represented in mature spores in horstail.These results proved that some proteins have been synthesized in mature spores before germination.Moreover,the abundances of various isoforms of 10 proteins such as RuBisCO large subunit,putative malate dehydrogenase,enolase,eIF4A-5,GTP-binding nuclear protein and dehydroascorbate reductase-like protein were detected in the analysis of differentially-expressed proteins during the germination of horsetail spores,respectively.This implied the various expressed pattern of these isoforms played great roles upon spores germination.
