| Two benthic diatom Navicula. Corymbosa and Nitzschia sp. which was separated from DALIAN sea area was used as experimental material. Growth conditions and biochemical compositions of the two marine benthic diatoms would be researched. The effects of photoperiod and nutrient factors of the culture solution, such as nitrogen, phosphorus,iron, silicon, manganese, copper, zinc, molybdenum, cobalt,vitamins B1 and B12 on the growth of Navicula. Corymbosa and Nitzschia sp.were studied using Single and Orthogonal Design. The main results are summarized as following:The specific growth rate of two benthic diatoms could maximat when the photoperiod was 12: 12.Using Single Design, the specific growth rate of Navicula. Corymbosa could be maximum when the concentration of nitrogen, phosphorus,iron and silicon, manganese, copper, zinc, molybdenum and cobalt,vitamins B1 and B12 was respective 49.28,1.137,1.96 and 22.951 mg/L,1.5,0.1,0.1-0.2,0.1 and 0.5μmol/L,0.5mg/L and 4.0μg/L. The optimum medium of Navicula. Corymbosa was decided using Orthogonal Design: NaNO3 299.2mg/L, NaH2PO4 5.26mg/L, FeC6H5O7·5H2O 14.73 mg/L, Na2SiO3 150mg/L, MnCl2·4H2O 396μg/L, CuSO4·5H2O 17.5μg/L, ZnSO4·7H2O 28.7μg/L, Na2MoO4·2H2O 7.26μg/L, CoCl2·2H2O 116.2μg/L, VB11mg/L, VB128μg/L。Using Single Design ,the specific growth rate of Nitzschia sp. could be maximum when the concentration of manganese, copper, zinc, molybdenum and cobalt,vitamins B1 and B12was respective 0.5-1.0,0.08-0.1,0.3-0.4,0.06 and 0.2μmol/L,1.0 mg/L and 2.0μg/L.The optimum medium of Nitzschia sp. was decided using Orthogonal Design: NaNO3 74.8mg/L, NaH2PO4 11.864mg/L, FeC6H5O7·5H2O 2.88mg/L, Na2SiO3 100mg/L , MnCl2·4H2O 79.2μg/L, CuSO4·5H2O 20μg/L, ZnSO4·7H2O 86.1μg/L, Na2MoO4·2H2O 14.52μg/L, CoCl2·2H2O 33.2μg/L, VB1 0.3mg/L, VB124μg/L。The change of chlorophyll a was as same as the growth of two benthic diatom ,while other biochemical composition was varies with the diatom,but they achieved maximum under opimum condition. |
