Human alpha-lactalbumin(α-LA)is a small,acidic,Ca2+binding protein, which performs many physiological and health functions, for instance,α-LA can improve the development of babies' nerves, induce their sleep and stimulate their immunity, particularly, recent studies have shown thatα-LA has the ability of killing the bacteria and inhibiting the tumor cells. An experiment was carried out in order to study the expression of theα-LA gene in transgenic tomato plants and the results were shown as follows:First, the plant expression vector pBRALA was constructed, which contained all elements for plant expression and was suitable for transformation via Agrobacterium, such as CaMV35S promoter, both left and right border sequence for T-DNA in Agrobacteria, plant reporter gene gus, plant screening marker gene hpt.Second, the plasmid pBRALA was introduced into Agrobacterium tumefacicens LBA4404.Third,the tomato regeneration system was established and the influence of some factors on tomato regeneration was also studied using tomato cotyledons and hypocotyls.Fourth,α-LA gene was transformed into tomato plants via leaf discs and the 18 transformed tomato plants were obtained after screening. The PCR analysis and GUS staining were performed and the results indicated that theα-LA gene was introduced into 12 transformed tomato plants. The transgenic tomato plants grew, blossomed and bore fruits.
