In recent years,with the development of the life science research,people's research on organism also from organ,tissue to cell and subcell,the new test technology of chemical and biology have do a great contribution for the research of biology.Fiber optical fluorescence nanosensor have great advantage:litter volume,higher resolution,shorter response time,and less sample.As a result,the living cell could be detected with less damage,micromation, multi-parameter,real time and noninvasion.It's not only could be used for clinic diagnose, detection and protection for the living and working environment,and acquire the information from man-trap,but also have great application on the dynamic test of vivo biology cell with minimally invasive and real time.The p-2000 microelectrode puller were used for fabricate optical fiber nanoprobe,then coating the fiber with vacuum coating machine,finally, constructing a novel fluorescence nanosensor system with the coated optical fiber and fluorescein,and applied it to study the pH_i of single CA16 cell.The results showed that FIR was positively correlated to pH_i range from 6.2 to 7.9.Calibration curve equation was Y=-25.27826+4.48026X(r=0.99851,P<0.0001),the pH_i is 7.21±0.05.
