Salinomycin is a kind of polyether antibiotics which has been widely used in the world as coccidiostat because of its safety,high efficiency, broad spectrum and low residue.It is very important to enhance yield and reduce cost of production by screening of Salinomycin high-producing strain and optimizing of production technology.It was studied that rational screening of Salinomycin high-producing strain from F2-1#,and optimization of production technology.Firstly,rational screening of Salinomycin high-producing strain was studied by EMS and UV.The mutants 3-233#,3-244# and 3-250# was obtained,which potency was increased by 34%,44.9%和39% respectively,compared with the original strain F2-1#.Secondly,it was showed that the product of Salinomycin was effected by butyric acid.The high potency of Salinomycin was 40195.8 U/mL by the Salinomycin producing strain in shake flask with 0.24%butyric acid in slant. Thirdly,Optimization of culture medium was studied by response surface method(RSM).The results showed that the most important components to the product of Salinomycin were soybean cake powder 23g/L and industrial peptone 9g/L.Under the optimizing medium the yield of salinomycin was 60750μg/mL,increased by 33.9%compared with that initial conditions.Lastly,parameter correlation analysis of process in the early period of salinomycin fermentation was studied,which could give a reference basis to production and application of fermentation scale up.
