Estrogen is synthesized by granulose cells under the control of follicle stimulating hormone(FSH) and luternizing hormone (LH). Estrogen through its corresponding estrogen receptor(ERα,ERβ) to activation of the activity of receptor, transcriptional regulation as a factor to regulate the transcription of target gene expression,thereby it play the physiological function. These study shows that the the expression of ERmRNA in preimplantation embryo,it can only illustrate that ER gene was transcripited, but it is not reportde that the expression of ER protein on development of preimplantation embryo, especially in vitro (lack of estrogen). The expression of their phosphorylated estrogen receptor and distribution has not yet been a systematic in-depth study.Our research object is early embryo of mice in vitro, the density 7.5μmol∕L of 17β–E2 was joined into the KSOM culture medium to culture early embryo. we used laser confocal microscope to detect the expression and regulation of estrogen receptor ERαand ERβ, and their phosphorylated estrogen receptor before and after jioning E2on development preimplation embryo in vitro.The results:1. The expreesion of ERαis weak in the normal development of embryo in the 2–cell and 4–cell stage. ERαwas not detectable at the 8–cell and morula stage, at blastocyst, ERαhas a high level expression. After joining 7.5μmol∕L of 17β–E2,compared with the expression of ERαin the normal of embryos development, has undergone an obvious change, in particular the 8– cells and morula stage, the change is most notable.2. The expreesion of ERβis weak in the normal development of embryo in the 2–cell and 4–cell stage. ERβwas not detectable at the 8–cell and morula stage, at blastocyst, ERαhas a high level expression. After treated by 7.5μmol∕L of 17β–E2,compared with the expression of ERβin the normal of embryos development, has undergone an obvious change, in particular the 8– cells and morula stage, the change is most notable.3. Phospho-ERαis expressed in the normal embryonic development of various stage, but it is strongest expression in the blastocyst stage. We detected the expression of phospho-ERαtreated by 7.5μmol∕L of 17β–E2 from 2 - cells to blastocyst stage, and it was highest from 8 - cells to the blastocyst than normal of the expression.4. Phospho- ERβis expressed in the normal embryonic development of every stage, it was expressed gradually strengthened. it was not much change that expression of phospho-ERβtreated by 7.5μmol∕L of 17β–E2 of compared to the previous early that normal development of embryos.These results show that the expression of ERαand ERβwere detected on development of mouse preimplantation embryo in vitro, that estrogen may play biology effect through the two receptor. It has some certain effect on the development and implantation of blastocyst that the high level expression of phosphorylation of both in the blastocyst stage; It was regulated by estrogen levels that the expression of ER and their phosphorylation form in early mice embryo development .
