Lycoris is valuable as an ornamental,and is also an important material used in medicine industry. Up to now, many scholars have studied on Lycoris, which have covered many aspects. In this paper, the expressed sequence tags(EST) and the leaves development of Lycoris longituba were studied, with the observation of leaf growth,mining EST-SSRs from EST data, analysis of codon usage bias, mRNA temporal and spatial expression of auxin bingding protein. The main contents were as follows:(1) The growth mode and structure of different leaf segment of Lycoris longituba were studied. In this chapter, the methods of drawing equal distance lines and paraffin slice were used. The results were that the leaf elongate after coming out to the day mainly depended on the stretch of its base, and its width and thickness changed little.The structure of leaf indicates that the cuticle layer that covers on pyidermis is more infertile,the pyidermis only has one layer of cell,the mesophyll are composed by palisade tissue,spongy tissue and secretory tube.The leaf type near the upper segment is isobilateral leaf.(2) Searching on the Simple sequence repeats(SSR) from Lycoris longitub expressed sequence tags(EST) and primers designment.Make use of SSRIT analysis procedure,it was found that 345 SSRs in the EST of Lycoris longitub and there are 50 high polymorphic EST-SSRs among them. By means of the software primer 3, we have designed 30 pairs primer for electing.(3) Codon usage bias was studied by means of on-line analysis procedure: Codon W on Lycoris longituba, Arabidopsis thaliana,Zea mays and Solanum lycopersicum.12 condons have higher usage frequency and 11 have lower in all the 4 kinds of plants. Lycoris longituba has its own higher frequent condon: AGT, AAA, ACT,TGC, TAA,TTA.(4) Character analysis and structure forecast for auxin binding protein have been done by software Prot-Param,antheprot,ProtScale,PHD,Finger-PRINTScan and SWISS-MODEL.Auxin binding protein is the acditic protein type,and the physics chemistry character,primary structrue and high-grade structure of it between different kinds of plants are in a high degree similarity.(5) mRNA temporal and spatial expression of auxin bingding protein on the leaf of Lycoris longituba was indicated by in situ hybridization mthod. Expressing amounts of ABP mRNA on the upper leaf segment is higher then the lower segment. Distribution area of ABP mRNA on the upper leaf segment is also bigger than the lower.On the base of the leaf, the detective signal is so weak that they are no differentiates between in the leaf border and the bundle.
