Sporocytophaga can decompose cellulose, starch,protein,fat,grease macro molcule,they can produce exopolysaccharide which have some biology function such as immunity reinforcing activity and so on,it is important microbe resoreces..In days of old we carry through research of Sporocytophaga degradation cellulose.But the characteristic of they degradate starch produce polysaccharide and amylase enzymology is not reported .The purpose of the paper is orientate of amylase as well as optimize of amylase ,and primary measure yield of exopolysaccharide,discuss the process of degradate of starch.The reasult of experiment as follows:1 Both Sporocytophaga sp. JL-01and SD-01 can degradate starch produce amylose,as a result of coparative , JL-01 strain growth more soon than SD-01, amylose yield more high.We optimize the condition of JL-01 produce amylose, use peptone is nitrogen source,inoculum size 1%,pH is 7,bring up 48 hours ,the yield of amylose is tiptop,144hours amylose almost is 0.The quantity of is 0.25mg/ml.2 JL-01 and SD-01 can growth in basic starch medium and produce amylase , adopt liquid fermentation and centrifuge,we get settle of upside and precipition.measure their enzyme activity .The deposition of thalli is broken up by ultrasonic.The result indicate that their amylase all are outside of cell,the activity of inside cell is low.3 By means of optimize of medium,we found JL-01 and SD-01 can growth in acidity condition,the furthest adaption pH are 4.We adopt fluid fermentation,confirm temperature,PH,carbon source,nitrogen source and so on act on Sporocytophaga and had some impact on amylase.The result indicate , JL-01 strain original PH is 4,the temperature of rocking bed is 32℃,quantity of starch is 0.5%,the major carbon source is urea,rocking bed is 36h,the enzyme activity achieve pinnacle. The temperature of rocking bed of SD-01 is 30℃,other cultivation contion is the same as JL-01.The activity of JL-01 is more high than SD-01.4 we take the study of amylase character,ultrafitration gruff enzyme,salt out, diallysis, Sephadex G-100 gel chromatography and freeze drying,PAGE,we get single amylase groupThe analysis indicate,the molecular weight of the enzyme is 61000Da,the most right reaction temperature is 60℃, pH is 4.In 40-60℃, pH4.0-6.0 condition is relatively stabilization.
