OBJECTIVE:To study the distribution of phospholipase A2(PLA2) M-type receptor mRNA in the organs and tissures of rats.To determine the sequence of the PLA2 M-type receptor cDNA and analyze its evolution characteristic so as to provide necessary information to understand the biological effects and relationship between PLA2 and its receptor.METHOD:1.Total RNA was extracted by Trizol from the rats organs and tissures that each weighted 50-100mg,RT-PCR was performed to amplify the cDNA of sPLA2-â…¡A,sPLA2-â… B and PLA2 M-type receptor;2.The products were conducted by electrophoresis and followed by semi-quantity analysis;3. The products were engaged in gene sequencing and sequence analysis.RESULT:1.High quality of product was obtained after proper condition of amplify the cDNA of PLA2 M-type receptor;2.PLA2 M-type receptor mRNA presented in the tissue of kidney,testis,ovaries,uterus,spleen,brain,large intestine,appendix,stomach,liver,vessel,salivary gland,adrenal gland,prostate gland,lung.among them,testis and ovaries were the richest;3.We discovered that the presentation of PLA2 M-type receptor mRNA in brain,salivary gland, adrenal gland and prostate gland,trace in large intestine,appendix,and stomach; 4.Species differences was found in the distribution of PLA2 M-type receptor;5. PLA2 M-type receptor characterized by accelerated evolution,and was 99 percent in homology with the phospholipase A2 receptor(predicted)gene which was known.6.Gene sequencing and sequence analysis revealed that there was G412A mutant in different tisues between two rats.CONCLUSION:1.PLA2 M-type receptor mRNA are distributed extensively in rat organs,we first discovered that PLA2 M receptor mRNA exists in brain,salivary gland,adrenal gland and prostate gland,trace expression in large intestine,appendix,and stomach.2.There is obviously species differences. 3.PLA2 M-type receptor gene may evolve rapidly.
