19nt Myostatin gene is a negative regulator of muscle growth, it can inhibits muscle cell's proliferation. The mutation and deficiency of myostatin gene can cause the raise of quantity of muscle. The reason that the raise of quantity of muscle is proliferation of muscle cell and hypertrophy of muscle fiber. The raise of expression of myostatin can inhibit the synthesize of muscle protein, cause the declination of the quantity of muscle. Myostatin can inhibit the proliferation of muscle cell by the dependence of dosage and dependence of time, and its function is reversible. The prefabrication of DNA and the restrain of cell replication by the abduction of myostatin gene can injured the muscle cell's proliferation during catabolism illness and senescence procession. The study of myostatin gene is good for the cure of sweeny. The inhibition of myostatin gene is good for prevent and cure fat disease and sugar diabetes. Thy study on inhibition of muscle by myostatin gene is actively studied in recently years. In order to increase the quantity of muscle, the major research method abroad is gene knockout and transgene. In this research , the RNAi method is used to silence the expression of bovine myostatin gene. The recombinant plasmids targeting to bovine myostatin gene were constructed, put a fundamental steps for producing transgenic animals.1. 19nt interferencing sequences for selecting different positions of myostatin gene were designed for Sence -Loop -Antisence by using biology software after analysising bovine myostatin gene. There are 4 shRNA interferencing sequence and 1 HK were designed.2. Then, these sequence of chemical were annealed for double strands interference segment, which were made directional cloning to interferecing plasmid to construct recombinant plasmids GDF- 8-1, GDF-8-2, GDF-8-3, GDF-8-4, HK. The result indicated that segments were cloned rightly and theren't nucleotide insertion and nucleotide deletion, and recombinant shRNA interference plasmids targeting to bovine myostatin gene was successfully constructed .
