Biochip technology has received much more attention around the world in recent years. Biochip can be divided into two types including passive biochip and active biochip. The passive biochip is the most popular one used in research and clinic test presently. But this type of biochip has some disadvantages such as more subjective disturbances in the process of fabrication and detection; difficult to standardization; uncontrollable biochemical reaction condition and process; low efficiency; poor reproducibility and accuracy of the result. To solve these problems existing in passive biochip system, some active biochips have been developed. The active biochip is characterized by the features of rapidity, high-efficiency and automation, which is a basic element for the construction of lab-on-a-chip and the integration of bioassay processes.Hybridizing kinetics is an important aspect of nucleic acid chip techniques. The studies are helpful to design and optimize biochips, build up stable experimental system and to develop a quantitative method for experimental results.In this paper, we designed a novel active nucleic acid chip system, which introduces the external force and controlling devices to adjust the hybridization reaction on the nitrocellulose membrane.In this paper, we use hybridization basic principles to study the feature of hybridization on passive nucleic acid chip and build up the hybridizing kinetics model. On these bases, we study the novel features and influence factors of hybridization on active nucleic acid chip. Finally, we build up the hybridizing kinetics model of active nucleic acid chip. The results demonstrated that this active nucleic acid chip system is a rapid, stable and practicable system, which may enhance the efficiency of hybridization reaction and improve the repeatability and accuracy of nucleic acid chip analysis.Good foundations for further research have built and further investigations will be undertaken in the future.
