Building a metabolic reaction database of high quality for a microorganism is the basis of both quantitative analysis of metabolic network and melioration of bacteria species with metabolic engineering technologies as well as embodiment of systems biology wholly analyzing microorganism from microscopical point of view. The main reconstruction method is to complement and cure existing databases or reconstruct a new one by combining information from the Internet databases, text books, literature, patents as well as experimental facts.Bacillus subtilis is a gram positive model organism widely used in industrially and medical studies, and the study and improvement of this bacterial is becoming more. However, the incompletance of the knowledge about it inhibits the research much. Therefore,for the clearer understanding of this bacterical by the research and guiding the experiment, we aimed at Bacillus subtilis and collected genome annotation and metabolic reaction information from three authoritative Internet databases: KEGG, Subtilist and Expasy. Based on KEGG, we compared the BG no. and EC no. of the three databases with VBA to make the three databases complement each other by programming, and then we identified the acceptance or rejection of the information which we got from databases according to the KO from KEGG database and amount of literatures. Finally, we constructed a high quality metabolic network of Bacillus subtilis,and for the first time, we marked the data in the metabolic network by confidence degree to ensure its quality. On the basis of these work, we coordinated some special genes and reactions in the network for the succedent research and the expand of the metabolic network. Presently,we have 768 genes, 546 enzymes and 1084 reactions and 1592 gene-reaction relationships in our network, and we have preliminary finished the none-reaction gene, EC-uncertain gene, multi-substrate enzyme, general reaction transport reaction and subunit protein list.
