Objective To investigate the role of neural regulation in collateral vessel growth by studying the effect of denervation on expression of RAM11,intercellular adhesion molecule(ICAM),vascular cell adhesion molecule(VCAM)and Ki67 in the collateral vessels.Methods Ten Zealand white rabbits were anesthetized and operated by incision of the right sciatic nerve.Seven days later,the animals were anesthetized again,the right femoral nerve was incised,and the nerve governing the profound femoral artery was destroyed by dropping 10%phenol around the vessel.Then,acute bilateral femoral artery ligation was performed with two knots underneath the deep femoral artery,the simply ligated contralateral side was used as control. All the animals were allowed to survive for seven days then killed.The expression of RAM11(macrophage marker),ICAM,VCAM and Ki67(cell proliferation marker)in collateral vessels were detected by immunofluorescence with special antibodies against RAM11,ICAM, VCAM and Ki67.Resultsâ‘ In simply ligated-side collateral vessels,accumulation of RAM11 in the adventitia was evident.A few RAM11 positive cells were also observed in the media.In contrast,the number of RAM11 positive cells was lower in dennervation side collateral vessels which was scatted in the adventitia.â‘¡In simply ligated-side collateral vessels, expression of ICAM was significantly higher than that in femoral artery ligation plus dennervation side which was mainly expressed in the endothelial cells.â‘¢In simply ligated-side collateral vessels,the staining of VCAM was strong both in the endothelial cells and in the adventitia,In the collateral vessels from femoral artery ligation plus dennervation side, staining of VCAM was weak.â‘£In simply ligated-side collateral vessels, cell proliferation(ki67)was significantly increased which were present in all layers of the vascular wall;In the collateral vessels from femoral artery ligation plus dennervation side,Ki67 positive staining was rarely found,occasionaly in endothelial cells.Statistic analysis showed there was a significant difference about expression of RAM11,ICAM,VCAM and ki67 in collateral vessels between simply ligated-side,and femoral artery ligation plus dennervation side(P value less than 0.05)Coclusion Our data show that dennervation leads to a down regulation of RAM11,ICAM,VCAM and ki67 in collateral vessels, suggesting that neural regulation is involved in the formation of inflammation and plays an important role in collateral vessel growth.
