The hexose transporters of Saccharomyces cerevisiae are of importance to its growth and alcoholic fermentation. In this study, we focus on HXT7 gene which encodes a high-affinity hexose transporter hxt7p. Based on the principle of homologous recombination, the deletion was carried out by using the Cre/loxP system obtained from plasmid pUG6, with insertion the KanMX gene as a reporter. We also cloned the HXT7 gene from yeast strain ZS-13, transformed into the plasmid pUG6, and inserted a strong promoter PGK in front of the open reading frame of HXT7 gene, in this way we achieved the over-expression of HXT7 gene.Then growth and fermentation experiments were carried out, the growth curves and fermentation dynamics were measured, found that in the end of fermentation, the ZS-13 strain lacks HXT7 gene had a low hexose intake rate; and the ZS-13 strain with a over-expressed HXT7 gene incept more hexose. But the change of hexose intake amount has no significant influence on ethanol or glycerol yield, the metabolic flux research should be introduced in future studies, expecting to convert the extra hexose to ethanol as much as possible.
