Objective:Through studying the protein expression of HIF-1αunder the different hypoxic training pattern to Preliminarily discuss the promoting effect of HIF-1αon angiogenesis in cardiac tissue.Through micro image analysis and with the CD34 this special vein endothelial cell surface antigen to mark vein,from the level of increased angiogenesis of the cardiac tissue to study the different hypoxic training pattern has effect on angiogenesis.From the protein expression of hypoxic inducible factor-1αand angiogenesis to study the function and mechanism of hypoxic training in training.At the same time,it hope that this theory can provide experimental basis and applied methods for some related disease.Methods:(South central university Xiangya medical school animal department provided)60 male SD rats were randomly divided into 6 groups according to the weight,each group n=10:Control,Hypoxic group,Training group,Living Low Training High,Living High Training Low,Living High -Training High-Training Low.Combine the progressive treadmill exercise,progressive hypoxia housing and the specific hypoxia concentration(hypoxia concentration 17.5%,is equal to level of 1500m)training stimulation to establish animal model.After the experiment,takes the cardiac tissue to be fixed,to make wax specimen,with the application of immunohistochemical technology and computer image processing methods to analysis quantitatively to the HIF-1 positive expression;with the application of immunohistochemical technology,computer image processing methods and stereological method to check and count the cardiac tissue of capillary density,gray level,the area expression.Results:(1)Under the normoxic,hypoxia-inducible factor has weak expression.Under the hypoxia,compared with the control group, hypoxia-inducible factor in cardiac tissue were significantly increased(p<0.01).(2)Hypoxic training groups compared with training group, hypoxia-inducible factor in cardiac tissue expression was significantly increased in the hypoxic training groups(p<0.01).(3)Pure hypoxia cannot increase obviously capillaries density in the cardiac tissue of rats.But the training and hypoxic training is able to increase the capillaries density(p<0.01).(4)Hypoxic training groups compared with training group, Capillaries density in the cardiac tissue of rats is increased significantly in the hypoxic trainings(p<0.05). Conclusions:(1)Regardless of which kind of be lack of oxygen,including the envionment and sport hypoxia,the hypoxia inducible factor-1αwill be increased in the cardiac tissue.Compare the sport hypoxia with envionment hypoxia,the sport hypoxia has the positive function.(2)The protein expression of hypoxia inducible factor-1αis remarkable in the cardiac tissue of rats if hypoxia combine with training,because the hypoxic training promote easily the expression of hypoxia inducible factor-1αthan pure training or hypoxia.and the livinghigh-traininghigh -traininglow group is the best way to promote the expression of hypoxia inducible factor-1αin the cardiac tissue of rats.(3)Pure hypoxia cannot increase obviously capillaries density in the cardiac tissue of rats.But the training and hypoxic training is able to increase the capillaries density.(4)The hopoxic training can promote easily angiogenesis compare with the training purely in the cardiac tissue of rats.which shows hypoxia with training together or training with hypoxic stimulus will have more positive effect on angiogenesis,and the livinghigh-traininghigh -traininglow group is the best way to promote angiogenesis in the cardiac tissue of rats.(5)Hypoxia inducible factor-1αis positive related with angiogenesis which shows that the HIF-1αhas the key fuction to promote angiogenesis in the cardiac tissue of rats,but the reason for the HIF-1αcause the expression of VEGF is unknown.
