Chromosome condensation is a significant process of cell multiplication and occurs as a prelude to sister chromatid segregation that initiates at the onset of anaphase. Structural maintenance of chromosome proteins, formed by two SMC subunits and three non-SMC subunits, was discovered in the 1990s. It's intergrant of chromosome and takes part in chromosome condensation and chromatid segregation. With the purpose of further study of the influence of condensin to chromosome condensation, we set up the eukaryotic expression plasmid cotransfection system which is tagged by GFP. We use the RNAi plasmids RHE and RHC which are constructed in our laboratory and idio-interfere the SMC subunits of cendensin to knock down the quantity of hCAP-E and hCAP-C of HeLa by RNAi technic. We find that chromosome condensation is exceptional, cell division is imcompletely, chromatin bridge appearant in the cells and also some cells which have more than one nucleus appear when the SMC protein expression is knock down , but other proteins are constant. These consequences prove that the two SMC-subunits play important roles in the process of chromosome condensation, not a single one can be omitted, they are the indispensable protein to chromosome condensation. This experiment has essential biology significance that we first adopt the cotransfection system and RNAi technic to study the hunman condensin SMC subunits in this experiment. The consequences we get provide valuable informations for the deeply study of the mechanisms of chromosome condensation and the condensin which takes part in chromosome condensation.
