Based on ten populations of Allium mongolicum, this paper assesses the genetic diversity among all populations selected, and that among different sub-populations within one population, which only involves 4 of total ten populations. The method of CTAB is used to obtain the genome DNA which is amplified by RAPD. It indicates that the Shannon index of Allium mongolicum is 0.2977 among ten different populations and the genetic differentiation index is 0.0174 among different populations, that is, 1.74% genetic variation is among populations while 98.26% is within one population. The index of gene flow is 28.235 among different populations, which may relate to their more extensive distribution. There is no interrelation between genetic distance and geographic distance of different populations.Four populations of Allium mongolicum are selected for illustrating the genetic diversity among sub-populations of each population. It reveals that Shannon index is 0.3447 among sub-populations within one population, showing rich genetic diversity of Allium mongolicum. The index of gene flow is 1.8650 among different sub-populations within one population. It may relate to its mechanism of pollination and scattering seed. The genetic distance correlates to geographic distance among sub-populations of each population. Of the four populations, that of Left Alashan Banner population shows the richest genetic diversity, whose index among sub-populations of population, the highest, is 0.2707, while the index of population in Saihantala, the lowest, is 0.2060, showing the poorest genetic diversity of this population, which is seen as the proof of the species'ability to adapt an arid environment and its evolutionary potential.Allium mongolicum have a rich genetic diversity, it has strong evolutionary potential and large adaptability to the change of environment, as a result, it is necessary to make reasonable utilization and conservation.
