Bioinformaton analysis of Nanog Oct4 Sox2 regulation mechanism for pluripotency of mouse embryonic stem cells.Following is the researching contents:1 Screening gene of the mouse embryonic stem cells which related to regulation of transcription By Nanog, Oct4, Sox2 target sequence. The result of analysis show that Nanog target genes Zbtb9, Shox2, Foxn2 Dmrt2; Oct4 target genes Ascl4, Uimc1, Arid2, Asb1; Sox2 target genes Zfy2, Tcea2, Tcfcp2 were study less and have the value of bioinformatics analysis.2 Through analysis of the physical properties and the secondary structure of the target genes show that Zbtb9, Uimc1, Zfy2 is transmembrane protein, Shox2, Foxn2, Dmrt2, Ascl4, Arid2, Asb1, Tcea2, Tcfcp2 is nuclear protein. These genes are rich in hydrophobic amino acids and has been the main helix.3 Through analysis of promoter region of target genes show that Nanog, Oct4, Sox2 the target sequences in the promoter region, or in the vicinity of the promoter region, or between the promoter region and the transcription initiation site, implied the transcription factor regulation the target gene by transcription. The most of target gene promoter region contains JCV and AP-2 binding sites showed that the target gene play an important role of the stem cells self-renewal. According to the function of (Sp1, NFI, H4TF1, junB) in the transcriptional regulation of specific cells predictable the target genes function, implied that Uimc1, Tcfcp2 play a role in nerve cells formation; Ascl4, Arid2, Foxn2, Dmrt2, Tcea2 play a role in in endothelial cells formation; Ascl4, Tcea2 play a role in muscle cells formation; Zfy2 play a role in keratinocyte formation.4 Nanog, Oct4 plays an important role of regulation in the differentiation of muscle cells and endothelial cells; The Sox2 play an important role in the differentiation of neural cells. The regulation of Nanog, Oct4, Sox2 have crosstalking. According to the Analysis established the regulation network for pluripotency of mouse embryonic stem cells.
