Many research have demonstrated that insects, which different from higher animals, don't have perfect and single immunity systems, and lack of B/T-lymphocyte. But they have strong adaptive capacity and defensive quality. Insects can produce a spectrum of antibacterial matters/peptides when infected with bacteria or have their body wall injured. These antibacterial proteins/peptides can kill bacteria and inhibit further infects. Antibacterial proteins/peptides have low molecular weights and perfect resistant to temperature treatment, hard to be hydrolyze and lack of immunogenic. Antibacterial proteins/peptides are now being one of the hot spot in insect immunology and molecular biology. More than 200 antibacterial peptides have been isolated from different insects.Orthoptera insect Oxya chinensis were first used in this research, after use different inducements induced the antibacterial matter of Oxya chinensis were isolated and analyzed through Sephadex G-50 gel filtration chromatogram, RT-HPLC and Tricine-SDS-PAGE. The results showed that both imagoe and nit of locust produce antibacterial matters after inducement, they were isolated twice through Sephadex G-50 gel filtration chromatogram. The imagoes sample RP-HPLC chromatography appeared 3 peaks on 2.09 min, 5.38 min and 13.57 min. On Tricine-SDS-PAGE electrophoretogram, imagoe sample in 97 kDa, 37 kDa places have 2 strips. In this paper, parts of physical and chemical nature of isolated antibacterial matters were also analyzed. Results demonstrated that The antibacterial spectrums of the antibacterial matter from imagoes and nit of locust only have inhibit effects on Gram positive bacteria. They all have perfect resistant to temperature treatment (40℃-60℃) and extreme pH(pH4-10).Preliminary research was carried out in this article; our results further increased the category of antibacterial peptides, and paved the way for further analysis of this kind of antibacterial peptides.
