Recent studies indicated that nuclear actin has been implicated in diverse activities, including chromatin remodeling, mRNA processing and export, and the transcription mediated by RNA polymeraseâ… ,â…¡andâ…¡. Earlier work in our lab have demonstrated that nuclear actin could downregulate the transcription of p21, which is mediated by the complex, including nuclear actin and p53. To study the mechanism of the interaction between actin and p53, GST-actin and three deletion mutants were constructed, and then GST-pull down assay was performed upon the series of GST-actin and nuclear extreact transfected with p53. the results showed that the 1-18 amino acids on the N-terminal and the 338-375 amino acids on the C- terminal of actin play an important role in the interaction between actin and p53. GST-pull down assay was also carried out upon the series GST-p53 and nuclear extract transfected with actin, and we found the 1-42 amino acids on the N-terminal of p53 is necessary for the interaction. Furthermore, the results of GST-pull down assay also showed GST-p53 and His-actin could interact directly in vitro.
