PKR is a dsRNA-dependent protein kinase that is induced by interferon treatment. PKR is an important component of antiviral defense mechanism that belongs to the serine-threonine protein kinase superfamily and the eIF-2αkinase family. PKR was found in almost all mammalian cells. A fish PKR-like gene, named CaPKR-like, was the first identified fish gene most similar to mammalian PKR.In order to realize the expression of CaPKR-like in the tissues of non-mammalian, this paper used crucian carp and grass carp for the organization of biological material to study non-mammalian organizations in the expression CaPKR-like activity. Multiclonal antibody was prepaired from rabbit immunized by purified PZα1Zα2. Western blots revealed that crucian carp and grass carp organization (brain, spleen, liver, heart, muscle, kidney, intestines, skin) without induced, CaPKR-like had a very low level of constitutive expression. But the level was up-regulated in stimulated with Poly I:C for seven days in eight tissues of crucian carp and grass carp. In the heart, liver, spleen, kidney and muscle tissue of Crucian carp ,CaPKR-like expression is strong and is slightly weak in intestinal and skin. In the brain, heart, muscle, liver, skin tissue of the grass carp ,CaPKR-like expression is strong and is slightly weak in the spleen, intestine, kidney.Induced crucian carp by PolyI:C, then analyzed the tissues of crucian carp by immunofluoresence to understand the location of CaPKR-like in cells of the crucian carp. The results showed that the expression of PKR gene in the crucian carp organizations induced by Poly I:C for a week is stronger than it in the crucian carp without inducing tissues, and found CaPKR-like mainly localized in the cytoplasm of the crucian carp's cells.
