DNA, RNA are two basic nucleic acids, DNA, RNA recognition reagents or artificial nucleases are a new challenging filed that is quickly developing in chemical biology. The macrocyclic polyamines are a kind of excellent ligands and have a substantially high binding affinity with transition metal ions. Macrocyclic polyamine metal complexes as enzyme models are widely used in chemical biology, medicine and gene therapy. 1, 4, 7, 10-tetrazacyclododecane (cyclen) is a representative macrocyclic polyamines compound and shows high binding affinity with transition metal ions, such as Zn(II), Cu(II) and Co(II). Their metal complexes especially multinuclear complexes were widely used in DNA, RNA recognition.In this dissertation, dinuclear macrocyclic polyamines with an imidozalium salt group and their Zn(II) complexes have been designed and synthesized, DNA protection have been studied. A series of useful and creative results have been obtained.All new compounds were characterized by IR, MS , 1H NMR 13C NMR , and elemental analysis. The synthetic methods and effect factors of the title compounds have been systematically studied. A series of convenient and efficient reactions have been developed.Imidazolium ionic liquids containing dinuclear cyclen moiety and their Zn (II) complexes interaction with DNA was detected by the method of agarose gel electrophoresis and fluorescence quenching. The results showed that DNA could be protected efficiently by the ligands and their Zn(II) complexes. Moreover, Zn(II) complexes were much more active than the free ligands. It will be useful in DNA separation, purification, and detection, and possibly in genetic engineering and gene therapy.
