Density, concentration, temperature, and stress are theelementary variables of chemical and physical processes. Theyare normally characterized by a three-dimensional (3-D)distribution in space and are also variable with time. Methodsof obtaining such 3-D distributions as well as their evolutionwith time are always of concern for fundamental measurements.The refractive index (RI) is an elementary physical parameterthat is used to characterize the properties of opticallytransparent substances.Firstly, the author presents the research situation of thedomestic and abroad optical system for measuring refractiveindices of the medium, some of which can measure thickness ofmedium and others can measure refractive indices of medium.Although some of them may simultaneously measure refractiveindex and thickness, they measure only a layer. Therefore, wepropose a novel non-contact measurement method based onconfocal microscopy and high coherence interferometer-y. Meanwhile, with this method, we can measure not only therefractive index of uneven medium, but also the refractive indexof every part in the medium, ln addition, with this method we canlocate the exact position of the test point.During the thesis, I designed four optical path drawingof the measuring system, including the new system of opticalfilter. After design job, I searched equipments which we needed.Most of them are existing in the laboratory, and some bypurchase, some we made by ourselves. Then I completed the wholesystem, and made experiment of several samples.At last I make some suggestions on the improvement of thismeasuring system.
