Cryptochromes are blue light receptors that regulate various photomorphogenesis responses in plants, including inhibition of hypocotyls elongation, stimulation of cotyledon expansion, and regulation of flowering time. The functions of cryptochromes are more and more clearly in recent years. So far, there are three cryptochrome genes, cry1,cry2 and cry3, were discovered recently. As more and more functions of the cryptochromes were uncovered, we are encouraged to know what genes were regulated by the cryptochromes, because the cryptochromes are so important in plants. It is necessary for us to discover the cryptochrome how to regulated plant flowering by constructing mutants in the background of cry1cry2 double mutant. The SCC means the suppressor of CRY1 CRY2.SCC17-D which is early flowering mutant is one of the SCCs mutants. SCCs are transformed with the plasmid of pSK1015 that harbors four CaMV35S enhancers. The SCC17-D has early flowering phenotype so that it was a good mutant for us to discover the reason of early flowering time.This experiment focuses on the early flowering time of SCC17-D mutant, because the active T-DNA insert is at nearby of the COL16 gene, COL16 is one of the CO family genes, the CO is the key gene regulating flowering in photoperiod pathway, so we think the gene COL16 may involve in flowering in Arabidopsis.Brachypodium distachyon Beauv is one of the Pooideae grasses. BD21 is one gnotype of the Brachypodium distachyon (L.)Beauv, it is quite similar to Arabidopsis in stature, the size of genome, growth condition and life cycle, and has main characters of grasses, for example, the sensitivity to pathogens, and caryopsis. BD21 should be as one of the best model plants for grasses functional genomics. BD21 is one of Brachypodium distachyon Beauv. To discrible the modality of the BD21 will give us a very impotant clew for the advance researching.What I have done is as the following:1.The expression level of genes near the active T-DNA tag was detected.2.The genes which near the active T-DNA tag were cloned.3.To transform the Arabidopsis with these genes and got the transgenic plant. 4. Constructing the RNAi vector of COL6,COL7,COL8,COL16 and got the transgenic plant5.To analyse the over expression transgenic pant and RNAi transgenic plant, show that the early flowering phynotype does not simply caused by one gene6.PFT (PHYTOCHROME AND FLOWREING TIME 1) and FT(FLOWERING LOCUS T)expression level in scc17-D are raising7.BD21 is sensitive to photoperiod and perceived by leaves; it will take out the spikelets about 28 days after it burgeoning; and the apical meristem will developmet from the seventh day when it burgeon..
