In this article, a breeding strategy combining a general mutagenesis is carried out to improve Coenzyme Q10 yield of Rhodobacter Sphaeroides.To focus on the optimization of Rhodobacter Sphaeroides culture conditions, a systematic method is established to optimize the production of Coenzyme Q10 and the biomass.The production of Coenzyme Q10 of the strain in liquid state fermentation is 231 % higher than original culture conditions.To mutate Rhodobacter Sphaeroides by means of mutagenesis with diethyl sulfate(DES), ultraviolet ray(UV) and lithium chloride(LiCl) consecutively, 14 Coenzyme Q10 highly producting mutants are screened from Rhodobacter Sphaeroides by mutagenses and high through screening, and Coenzyme Q10 production of mutants named X9 in liquid state fermentation is 323% higher than parent strain.A simple method of extracting and determining Coenzyme Q10 content of strain is obtained. A high through screening established is a rapidlly screening method. They could be efficient methods for quantitative analysis of fermentation production of Coenzyme Q10. Finally, the inherited stability of the mutant strain is studied.
