At present, research on bone marrow mesenchymal stem cells (BM MSCs) is a hot topic in cell biology. A major area in regenerative medicine is the application of stem cells in bone reconstruction and bone tissue engineering. This will require well-defined and efficient protocols for directing the differentiation of stem cells into the osteogenic lineage, followed by their selective purification and proliferation in vitro. Stem cells for osteoregeneration therapy can be derived from two major sources: embryonic stem (ES) cells derived from the inner cell mass of blastocyst stage embryos(10,11) and mesenchymal stem cells (BM MSCs) derived from the bone marrow. In contrast to ES, MSC have many advantages,for exmamples, MSC is no need to overcome immunological rejection from the transplant recipient, and they can avoid the formation of teratoma, ect. However, the putative MSCs from bone marrow are in fact a highly heterogenous population, with only a very limited proportion of cells being capable of differentiation into the osteogenic lineage, and the differentiation efficacy is not high enough. To establish an effective method for inducing mouse bone marrow mesenchymal stem cells to differentiate into osteoblasts, we study the role of DNA hypomethylation in osteogenesis. DNA methylation is one of the general mechanism of controlling tissue-specific gene expression. The cytidine analog, 5-azacytidine, which does not undergo methylation at the 5-position, acts as a demethylating agent during cell growth. Thus, Treatment with 5-azac can cause DNA hypomethylation. The pretreatment of 5-azac before induction of differentiation showed stimulatory effects on osteogenic differentiation of mouse BM-MSC. We hope from this study, we can promote the application of stem cells for osteoregeneration therapy.
