Cellulase is widely used in medicine, heredity, textile, chemical industry for daily use, papermaking, environment, sources of energy, food fermentation, petroleum exploitation, resources reproduction, waste water handle, agriculture, animal nutrition, chinses herbal medicine effective component extraction,and so on. Cellulase application prospect will be a vast world. The screening for the strains with high cellulase activity and the studies on how to improve strains cellulase activity is very important for ecology,economy and our society.The samples selected from the campus and the forest area at Xinglong Mountain in Yuzhong County are firstly screened with Congo red cellulose identification culture medium and CMCNa culture medium. By this means, we select 160 strains with larger Hc value ( ratio of diameter of clearing zones and colony). Secondly, the 160 strains are fermented and we measure enzyme solution CMCase activity and FPA activity. At last, the 15 strains with higher CMCase activity and FPA activity are screened. Among the 15 strains with high cellulase activity, the cellulase activity of the strain 58-3A, the strain 42-2A and the strain 50-2B is the highest. The CMCase activity of the strain 58-3A is 1376.2U and the FPA of the strain 58-3A is 497.78U. The CMCase activity of the strain 42-2A is 1378.64U and the FPA of the strain 42-2A is 338.56U. The CMCase activity of the strain 50-2B is 1378.64U and the FPA of the strain 50-2B is 338.56U. They make filter paper rot rapidly. Observing these strains morpha, we find there are many kinds of bacterias, fungi and actinomycetes and some unknown strains among the 160 strains. There are filamentous fungi and actinomycetes among the 15 strains with high activity cellulase.The strain 58-3A is Penicillium LK. ex Fries. The strain 42-2A is Trichoderma Pers.ex Fr. The strain 50-2B is Rhizopus Ehrenberg.In future,the CMCase activity and FPA activity of the 15 strains with high activity cellulase can be improved by optimizing submerged fermentation. From now on, it is necessary that we should do some research of molecular biology for the calluase gene clone and the calluase gene sequence analysis of the strain 58-3A and the strain 42-2A.
