Slowness of cell upgrowth resulted from insufficient oxygen supply is an important limiting factor in large scale culture of plant cell. However, it is observed that VHb, which is a bacilli hemoglobin, not only be able to integrate with low density oxygen efficiently, but also can release oxygen quickly, moreover, it is profitable for utilization of low density oxygen. For the sake of studying whether VHb can be applied in plant cell culture, we have mensurated in this paper the enzyme activities on tobacco suspension cell lines corresponding to w1 and t12 and respiratory pathway of tobacco callus , such as alcohol dehydrogenase ( ADH ) glucose-6-phosphate dehydrogenase ( G6PD ) , phosphofructokinase (PFK) , pyruvate kinase (PK) , succinic dehydrogenase (SDH), isocitric dehydrogenase (IDH) and MTT assay. The results showed that in tobacco suspension cell lines, the enzyme activity of t12 was higher than w1 and the trends of enzyme activity about w1 and t12 both are similar to the growth trend of the cell biomass. Similarly, the above results also can be obtained in the study of enzyme activity on tobacco callus. In the mensurations of the two system, it both can be obtained that the enzyme activity of t12 was higher than w1, which show that VHb play a great role to improve the growth capacity of the tobacco suspension cell on t12, furthermore it can provide the theory for culture of plant cell in large scale.
