Bioactive peptides (BAPs) widely distribute in many living organisms. They have special. function such as anti-microorganism and anti-tumor. Recent researches of BAPs focus on the mechanism of anti-microorganism. Patch clamping has evolved many recording modes for different studies and became an important technique in modern hymenology and electrophysiology. Application of the patch clamp can deeply investigate in ion channel, cell membrane selectivity penetration and ion channel modulation mechanism et al.We use electrical stimulation to distill northeast forest frog (Rana dybowskii)skin secretion, and the secretion purified by Sephadex G-100,50,25,collect the exudation which is active object, Separate the exudation with semi-product HPLC and inspect in violet detector with wavelength is 216nm then get eleven kinds of BAPs.named dybowskiiⅠ~Ⅺ. Desiccate the BAPs with low temperature and negative pressure drier then got the pure BAPs power. Dissolve the pure BAPs power with suit solution and make the end density are 20ug/ml, 100ug/ml,500ug/ml.The result of perforated patch mode shown the BAPs which density are between 20ug/ml to 500ug/ml could pore the human breast cancer (MCF-7) cell membrane, during fifteen minuses after giga seal check the bore used method of electrophysiology. This effect is stronger depended on the raise of BAPs'density. But 500ug/ml BAPs didn't have effect on smooth muscle cells of rat in the same condition.The result of whole cell patch mode shown the BAPs could restrain the delayed rectifier kalium current of MCF-7 cell, the current density descended from 29.65±4.26pA/pF to 23.15±3.25pA/pF(n=5, p<0.05);the kalium peak current decline 21.93 percents. But BAPs didn't have effect on smooth muscle cells of rat in the same condition. And the peak current decline 0,03 percents (n=5, p>0.05).The whole Cell patch clamp also shown the BAPs restrain the solium current and make the current density descended from 110±1.26pA/pF to 80.15±9.65pA/pF(n=4, p<0.05); The solium peak current decline 27.26 percents. The dybowskiiⅠ, dybowskiiⅡ, dybowskiiⅢ, dybowskiiⅣcan also make the current density descended.Survey the voltage depended kalium and solium ion channls during BAPs effect on MCF 7 cell reflect the standed of Electrophysiologic,the result shown BAPs maybe could anti-tumor.
