In this thesis, the proteins of SARS-CoV and influenza virus were expressed byusing baculovirus expression system.Chapter 1 was a general introduction of expression systems, especially thebaculovirus expression system. SARS-CoV and influenza virus were also introduced.In Chapter 2, eight structure proteins including S, S1, S2, N, 3a, M, 7a, E ofSARS-CoV WH20 strains was expressed by using AcMNPV Bac-to-Bac expressionsystem and pFastBacSHTa. SDS-PAGE and Western blot analyses indicated that all theeight proteins were highly expressed and with good antigenicity. The expressed Eprotein was purified and used to generate a polyclonal antibody.In Chapter 3, ten proteins, including HA, NA, NP of several different influenzavirus strains were expressed by using AcMNPV Bac-to-Bac expression system and thetransfer vector of pFastBacSHTa.In Chapter 4, the surface display system of AcMNPV was introduced. Byinserting the eGFP and S of SARS-CoV WH20 strain in between of the signal peptideand the mature GP64 of AcMNPV, and the eGFP-S were expressed and integrated withGP64. Western blot analysis indicated that the eGPF-S were integrated into the surface of the cells.
