Objective: Aflation B1(AFB1) is one of the most potent toxic and carcinogenic substances. Armillariella sp(E-20), that produce aflatoxin-detoxifizyme(ADTZ), has been screened out by our research institute, and recombinate ADTZ(rADTZ) has been get from the method of genetic cloning and recombinate expression. Earier research showed that ADTZ detoxify AFB1 though hydrolysis reaction, following the oxidizing reaction. Oxidation is an important priming process. This paper uses electrochemical method to discuss the path of ADTZ catalyzing AFB1, and the character of ADTZ.Method: Ion exchange chromatography(IEC) and hydrophobic interaction chromatography(HIC) were used to purify and prepare high activity rADTZ and ADTZ. Nanomaterial CNTs with carboxyl group immobilise rADTZ and ADTZ by means of cross-linked. Eletrochemical methods, Cyclic voltammetry(CV) and Different pulse voltammetry(DPV), were employed to detect the oxidoreduction signals of Aflation B1(AFB1), Sterigmatocystin(ST), Versicolorin A(VA), 2,3-Dihyopynan(DHP), Tetrahydrofuran(THF), Benzofunan(BF) and 8-methoxy-psoralen(MOP). HRP was employed to comfirm the production of H2O2, and the influence of O2 on oxidoreduction signals during ADTZ catalyzing was inspected by bringing N2 or O2 to the system.Results:(1) AFB1, ST, VA, DHP, THF, 8-MOP and BF were detected by rADTZ-SWNT modified electroed, among which AFB1, ST, VA, DHP and THF all had redox signals while 8-MOP and BF had no significant signal.(2) The ADTZ-AFB1 reaction mixture was detected with HRP-SWNT modified electrode and resμlts showed that H2O2 were produced.(3) Oxygen enhanced the redox signals of the ADTZ-AFB1 reaction.(4) The resμlts showed oxidoreduction existing in the catalysis of rADTZ. The production of H2O2 and the enhancement of redox signals, under the influence of O2, rADTZ was supposed to be a kind of mono-oxygenase.
