Immunochip is a kind of protein chip. It can realize immunoassay by protein probes immobilized on the glass slide surface capturing labeled target protein with special immunological reaction. The chip image can be gotten by laser scanner and analyzed by special computer software. So there are many advantages of immunochip techniques, such as high throughout, automation, high sensitivity and multianalysis.Double antibody sandwich detection can't be realized without employing immunochips to two sites small molecules target object. Small molecules can be dectected by the competition pattern. The first step is to conjugate small molecules with protein and then labeling the protein by cy3. Five small molecules can be linked to immobilized antibody competitively, and then mix it with protein which has linked the small molecules, Then concentration of the analytes can be detected in the sample.In this experiment, five difference chemical reactions were carried out because of the structural difference between atrazine, nonylphenol, 17β-estradiol, paraverine and chloramphenicol. The linking with protein was successful, and the linked small molecules could be recognized by protein-chip. After conjugated small molecules with BSA or OVA, the immunochips were developed to detect five small molecules qualitatively and quantitatively in water sample. The result shows that fluorescence signal intensity increased with the decreasing concentration of analytes. It shows a linear trend from 0.001μg/mL to 5μg/mL. And the immunochip was developed successfully for the detection of five small molecules on the same chip.
