Backgroud: The gene of bone morphogenetic proteins receptor II (BMPR- II) encodes a transmenbrance serine /threonine kinase, It is the important member of the BMP-signaling pathway.It can regulate multiple cellular processes, including cell differentiation and migration. Mutations in BMPs can cause several diseases, such as skelltal defects, familial primary pulmonary hypertension and neoplasias. The BMPR-II gene has 13 exons. it can bring forth two transcript variants through selective splicing. The variant 2 lacks a 1280 nt exon within the coding region compared to variant 1, that is, the variant2 is absent of the corresponding 12nd exon sequence of the transcription variant 1. The state of 5' CpG island methylation is closely correlative with gene espression ,and it is commonly thought that DNA methylation is negatively correlated with gene espression. The gene espression weakens when the degree of methylation is high and vice versa, the gene espression enhances when the degree of methylation is low.Methods: We investigate the level of two transcription variants of BMPR- IIgene in three cell lines( Helf,PGCL3, NCIH460) by RT-PCR and Real-time PCR; We also check the degree of methylation of BMPR-II gene 5' CpG island of above four cell lines in order to find out the potential relationship between the degree of methylayion of BMPR-II gene and its espression .Results: The results of RT-PCR and Real-time PCR show that the expression level of two transcription variants of BMPR-II gene in the Helf cell was lower than that of other two cell lines The MSP results show that methylation of BMPR-II 5' CpG island was found in Helf cell line ,Nonmethylation was found in NCIH460 and PGCL3 .But in A549 neither methylation nor nonmethylation are detected.Conclusion: At a certain extent, The BMPR-II gene espression is correlated with the degree of BMPR-II gene 5' CpG island methylation.
