In order to verify the effect of recombine leptin reducing fat deposition and study the effect on endocrine metabolism,recombine leptin was firstly prepared in rat and the pattern of fat SD rats was founded in this experiment,which would afford theory evidence and experiment data for the therapy of adiposity and the improvement of growth performance and meat quality.This experiment chosed 75 SD rats,weighting approximately 50g~60 g.75 SD rats were allocated randomly to five groups(Fifteen rats in each).GroupⅠwas used as control group for founding the fat pattern and fed with lower energy feed,groupⅡ,Ⅲ,Ⅳ,Ⅴwere used as experiment group and fed with higher energy feed.After the fat pattern had founded,GroupⅡwas used as control group for leptin injection experiment and intraperitonealy injected with physiological brine,groupⅢ,Ⅳ,Ⅴwere used as experiment groups and injected with 1.0 mg,0.5 mg,0.1 mg recombine leptin,respectively.The result indicated that: After injecting fat patten rats with different quality recombine leptin,the content of fat metabolism index were higher than that of control group.Compared with control group,the content of HDL of experiment groups increased at different extent from the first week after immunized with recombine leptin,maintained to over the experiment and reached remarkable difference at three weeks(p<0.05);CHO,TG and LDL had downtrend from first week to second week after injected recombine leptin,began to go up three weeks later and maintained to over the experiment(p>0.05);GH were higher than that of control group first week later and maintained to the end of the experiment(p<0.01);The content of Ins began to drop after injected recombine leptin,reached the remarkable different at the end of first week and maintained the trend during the experiment;T3,T4 were higher than that of control group;IGF-1Ⅰincreased after injected recombine leptin,maintained to the end of three weeks and groupⅢ(high quality injecting group)were higher than that of control group (p<0.05).The experiment made it clear that recombine brine had important biological activity and physiological function,included feeling the nutrional state,restraining the food,stimulating or maintaining the wastage,effecting the endocrine system;Recombine leptin took effect on neuroendocrine system by contacting with GHRH,GHCH and CRH,restrained the nsecretion of Ins,promoted the increase of GH and IGF-Ⅰ,T3,T4,to promote the lipolysis of fat cells,effect the weight of subcutanceous adipose deposit.The experiment determination of biological activities of recombinant leptin in rats,The result showed that recombinant leptin could promote the lipolysis of fat cells decrease in content of triglyceride,Inhibition of food intake and decrease in body weight,established the base on the application of recombine leptin on obesity disease and animal production were satisfactory.
