The formation of pollen wall is important for the development of pollen, however, its mechanisms still remain unknown. We present here a comprehensive characterization of gene OsMS2 (MALE STERILITY 2) and demonstrate its biochemical function in the development of pollen wall. The osms2 mutant was obtained from 60Coγ-ray treatment. Genetic analysis indicated that the osms2 mutant male sterility phenotype was controlled by a single recessive nuclear locus. Using map-based cloning approach, the OsMS2 locus was mapped between the two InDel markers, SJ622 and CL7-4 on chromosome 3. The region delimited by these markerss was about 35kb. In this region, there was a gene homologous to Arabidopsis male-sterile gene MS2. The result of sequence analysis showed a deletion of one nucleotide in the eighth exon, which resulted in frame shifted mutation.
