| After consulted many articles about small molecule chemicals interact with DNA,the spectrophotometric characteristics of DNA complex with 4-(2-thiazolylazo)-1,3-naphthalenediol(TADNm),1-(5-bromo-2-pyridylazo)-2,7- naphthalenediol(5-Br-PADN),1-(2-pyridylazo)-2,7-dihydroxynaphthalene (2,7- PADN) and 5-(2-benzthiazolylazo)- rhodanine(BTARH) have been studied.1. The interaction of TADNm with DNA was studied. The optimum conditions for the reaction of DNA with TADNm were as following: pH4.1,1.2×10-4 mol·L-1 TADNm,1.5×10-4 mol·L-1 CTMAB,12℃and reaction time of 20 min. In the above condition, DNA forms an orange-red complex with TADNm and its maximum absorption peak is at 470 nm. Beer's law is obeyed in a range from 3.75 mg·L-1 to 17.5 mg·L-1. The proposed method is applied to determination of DNA in analog sample and the recycle rate is from 97.1% to 101%.2. The interaction of 5-Br-PADN with DNA was studied. The optimum conditions for the reaction of DNA with 5-Br-PADN were as following: pH9.5,1.0×10-4 mol·L-1 5-Br-PADN,1.5×10-4 mol·L-1 Triton X-100,room temperature and reaction time of 15 min. In the above condition,DNA forms an modena complex with 5-Br-PADN and its maximum absorption peak is at 580 nm. Beer's law is obeyed in a range from 4.75 mg·L-1 to 15.0 mg·L-1. The proposed method is applied to determination of DNA in analog sample and the recycle rate is from 97.2% to 103%.3. The interaction of 2,7- PADN with DNA was studied. The optimum conditions for the reaction of DNA with 2,7- PADN were as following: pH9.5,1.8×10-4 mol·L-1 2,7- PADN,1.5×10-4 mol·L-1 CTMAB,room temperature and reaction time of 20 min. In the above condition,DNA forms an kelly complex with 2,7- PADN and its maximum absorption peak is at 420 nm. Beer's law is obeyed in a range from 7.50 mg·L-1 to 50.0 mg·L-1. The proposed method is applied to determination of DNA in analog sample and the recycle rate is from 97.1% to 105%.4. The interaction of BTARH with DNA was studied. The optimum conditions for the reaction of DNA with BTARH were as following: pH3.8,1.6×10-4 mol·L-1 BTARH,room temperature and reaction time of 30 min. In the above condition,DNA forms an yellow complex with BTARH and its maximum absorption peak is at 390 nm. Beer's law is obeyed in a range from 1.25 mg·L-1 to 5.00 mg·L-1 with the detection limit of 1.02 mg·L-1. The proposed method is applied to determination of DNA in analog sample and the recycle rate is from 95.0% to 103%. |
