With the homozygotes of 13/17 Robertsonian translocation swine [2n=36,XY or XX,rob(13;17)], heterozygotes of 13/17 Robertsonian translocation swine [2n=37,XY or XX,rob(13;17)] and normal swine as samples, microdissection and microcloning were carried out with mitosis metaphase chromosome preparation which was prepared by peripheral blood lymphocyte.The microcloning library was constructed and positive clones were screened and analyzed.The peripheral blood lymphocyte was stimulated with ConA or PHA and the optimal concentration was studied. The results showed that the optimal concentration of ConA was 0.4mg/ml or 1mg/ml for PHA, and the effect of ConA stimulating was significantly better than that of PHA (t(0.05)18=2.101
