Background and Object Genomic imprinting is an epigenetic modification in the germ line leading to preferential express one of the two parental alleles.Studies have shown evidences that genomic imprinting play an important role in carcinogenesis and could be regard as molecular marker for some malignant tumor.CTCF(ccctc binding factor) is a highly conserved nuclear protein. CTCF has been identified localizing in chromosome segment 16q22, a region that is often deleted in tumors which led to the hypothesis that CTCF is a tumor suppressor.Defming the exact function of CTCF has proved challenging. This protein binds sites within promoters,silencers, and insulators through the use of different combinations of zinc fingers.This protein has been implicated in tumor suppression,gene regulation,cell growth and x-inactivation,et al. As a protein with such diverse functions, One might assume that CTCF would have an equally complex set of regulatory parameters.IGF2 is one of the imprinted genes in human genome.in our past study,we found CTCF played an important role in imprinting of IGF2, so we screened interaction protein of CTCF by yeast two hybrid system to pave the way to study these proteins' role in regulation mechanism of IGF2 imprinting and the way to recover imprinting of IGF2 gene.
